You've misunderstood what those buttons are for. They're one-shot Override buttons (added at users' request...) to change the default behavior of what the profile is set to do (that is, do what it says on the buttons instead, if different). If you have a Mirror Right profile already, then
a) it is probably suggesting the relevant actions already (and you just need to hit Continue)
b) clicking the 'Mirror all files to Destination/Right' button very likely would not change anything anyway (see 'a' above - it's probably already proposing exactly those actions)
But if you had a Backup profile, you could (on occasion / on demand) switch it to Mirroring temporarily (say, to delete orphans on Destination periodically), but without having to reconfigure the Decisions (and without having to remember to change them back again afterwards)
But clicking the alternate ('Mirror all files to Source/Left') certainly will change something - it will reverse the entire actions of the profile (and the grid above will change - often radically - to reflect that). Make sure you know what it will do (for example, press F1 and read the Help on the Differences window...)
