Use Backup or Mirror profiles (read the Help for the difference) for strictly one-way traffic unless you re-configure them otherwise.
You can use smaller (overall) drives for your backup (partitioned accordingly or just use segregating base folders in your profile's Destination so that backup of E:\ will go to (say) X:\E_Backup\ )
This will work unless and until the available space is exceeded by the amount of (selected) data on the relevant Source. Ther's nothing magic about that concept.
Basically, if Explorer can see it (by drive letter), so can SyncBack,
