If it's not listed under When > Insert as a hardware serial to choose from, the Windows API we use (AFAIK) is simply not detecting a serial for that drive (not reading the drive's property string correctly or failing to parse it properly, I guess). I seem to recall I had that issue with a previous drive and Belarc Advisor scan didn't list it either (or, it was blank, I forget exactly). Unfortunately I don't recall which drive it was or even if I still have it, and it's listing all my current drives either way.
I'll check with the development department if it is an API (I think so), but if you don't hear back, I was right (thus, nothing we can do; it might even be a simple minor incompatibility with your drive v. Windows)
Do note that the drive must have been inserted before you click the Run This Profile When checkbox in When > Insert. If in doubt, uncheck it, close and re-open the Modify window, then check it again to force a rescan of connected drives. But if it still doesn't work if you enter it manually, sounds like Windows simply isn't reading a value from the drive that matches.
For your 2nd question, if OnInsert is working (e.g. with ordinary SERIAL, Pro is obviously running, so just switch to it, right-click the profile and Run
