Yes. De/selections are not the same as Filters, and in fact override them.
Thus you can filter out all *.TXT but select (include) MyVeryImportantLog.txt for backup.
Or, you can filter in all *.TXT but de-select (exclude) MyHugeLog.TXT when (say) backing up to a small flash drive. In either example, the de/selection overrides the relevant Filter.
Adding new Filters (or editing them) instigates a re-scan of all your current filters against all your current de/selections, and proposes review of any mismatches it finds (which may be intentional, as above).
Adding new de/selections doesn't add new Filter entries, because de/selections trump Filters anyway.
