Q1) it's not 100% clear if you want to back up all files as-of a change to any one of them, but I think that's what you're asking for - if you do, set the Destination to include
....\%DATE%-%TIME%\
and set the profile to trigger OnChange in Source (Modify profile > Expert mode > When > Changes)
This will copy all files to the new (empty) Destination it just created for you, whenever any of them change. Bear in mind Pro has to be running all the time for the OnChange detection to work.
Note that for any schema which produces a non-cycling potentially-infinite backup series (like the above), you are responsible for monitoring available space, deleting old backups, and so on. There's no functionality to auto-delete them in the program, except by virtue of re-using locations specified by a Destination string that 'cycles' (e.g. days 1-7 / repeat)
Q2) use Modify profile > Expert mode > Misc > Eject (for Destination). This will perform a Safe Removal (flush the cache for the device). It obviously cannot actually physically eject (like it can for a floppy or CD), and will tell you so if you use Test, but the S/R has been done and the device will be disconnected.
Bear in mind that if you set (or Test) this, and it triggers, the device will have to be physically removed and re-connected before you can use it again.
There's no built-in notification this has happened (bear in mind that on a Scheduled run in Vista-and-later there would likely be nowhere to display it anyway). You can add your own script to Programs-After if you want (a simple VBScript with a MsgBox "OK to remove" would do - but remember the same issues apply on a Schedule as per above link), or you might want to consider using Misc > Speech for an audible alert (which would work even if run on a Schedule, if anyone was around to hear it)
